Would it be possible to allow annotation in white or yellow on the black background when using the spotlight tool?

Strictly speaking no. BUT there is always a work around!
Pull out a black square and right click on it so that you can alter its properties. Pull it to the top layer so that it can now be treated as an annotation.
The eraser tool can now be used to create your choice of spotlight shape. This erasing action is effectively creating a moveable see through annotation. This now means you have a spotlight effect, but more importantly you can write all over your black square with a yellow or white pen. The attached example is done in Primary but demonstrates it possibly more clearly than I have described it. There are some page notes saved there too.

There's another possibility too when you're feeling adventurous. If you go into Settings (Click on Promethean Man > Studio Settings) and then select Right Button. If you choose Start Camera Tool, and select the time delay you wish, you can take snapshots of spotlighted areas with as much or as little black showing as you want. Put them on another flipchart page, then annotate at will!
Do remember to turn that right button setting off again when you don't want to use it! :D
